Summary
This Administrative Supplement request aims to perform single nucleus gene expression studies (snGEX) on
white blood cells from buffy coat samples collected as part of the Acute to Chronic Pain Signatures (A2CPS)
program. snGEX will be assessed at two timepoints per individual: baseline and 3 months post knee
replacement surgery. Gene expression profiles from subjects who develop chronic pain will be compared to
profiles from those who do not.

Narrative
The goal of the Acute to Chronic Pain Signatures (A2CPS) program is to identify biomarkers and their
collective biosignatures that predict susceptibility or resilience to the development of chronic pain after an
acute pain event. In this administrative supplement, the UC San Diego Omics Data Generation Center (ODGC)
will use white blood cells (WBCs) from buffy coat samples collected by the A2CPS to determine single nucleus
gene expression signatures associated with the development of chronic pain. This project is relevant to public
health because it will aid in our understanding of the role of immune cells in chronic pain and complement
biosignatures developed by the parent grant.
